mRNA vaccines activate immune responses through unexpected pathways, providing design insights to improve future cancer vaccines.

Unlike conventional vaccines and infections that rely on cDC1-mediated cross-presentation, mRNA-LNP vaccines engage both cDC1 and cDC2 redundantly and leverage cross-dressing (peptide-MHC transfer from non-hematopoietic cells) in a type-I interferon-dependent manner. This mechanistic insight explains how mRNA vaccines can elicit CD8+ T cell responses to bystander antigens and provides a rationale for optimizing future mRNA cancer vaccine designs.
