Combining a BTK inhibitor with an antibody drug enhanced lymphoma killing, especially in a subtype traditionally resistant to BTK inhibitors.
Zanubrutinib (BTK inhibitor) synergized with polatuzumab vedotin (anti-CD79b ADC) in DLBCL by upregulating CD79b expression on cancer cells, enhancing ADC efficacy especially in GCB-subtype DLBCL — the historically BTK-inhibitor-resistant subtype. In vitro and in vivo data provide mechanistic rationale for a combination already being tested in clinical studies.
