This preclinical study tested an antibody-mimetic drug conjugate bearing the duocarmycin DNA alkylating payload combined with an ATR (ataxia telangiectasia and Rad3-related) DNA damage response inhibitor in a HER2-overexpressing breast cancer xenograft model. Complete tumor regression was achieved with the combination, providing preclinical proof-of-concept for pairing ADC-mediated DNA damage with ATR pathway inhibition to prevent repair.
