Association of GLP-1RA discontinuation and risk of depressive and anxiety disorders in people with type 2 diabetes: a cohort study.
Patients stopping certain diabetes drugs should watch for mood changes, a finding that helps prevent unexpected psychiatric risk.
Using the Shanghai Hospital Link Database, this study found that stopping GLP-1RA therapy in T2D patients increased the risk of developing depression or anxiety compared with stopping DPP4 inhibitors or SGLT2 inhibitors, despite GLP-1RAs showing neutral or favourable psychiatric safety during active use. The finding—mediated in part by triglyceride rebound—highlights a rebound psychiatric risk signal that warrants monitoring in the millions of patients who discontinue GLP-1RAs annually.

Why it surfaced
Novel pharmaco-vigilance finding with major public health implications given widespread GLP-1RA prescribing; Nature Metabolism publication; observational design limits causal inference so design_quality capped; active-comparator design and large real-world cohort strengthen validity.
