This study introduces antibody-oligonucleotide-drug conjugates (AODCs), a novel programmable ADC modality using noncoding oligonucleotide prodrug scaffolds to expand chemotherapeutic payload capacity while enabling targeted antigen-specific delivery. HER2- and CD38-targeting AODCs carrying gemcitabine prodrug chains demonstrate immunogenic cell death, dendritic cell activation, and enhanced anti-tumor efficacy in combination with PD-1 blockade, supporting AODCs as a generalized platform for chemo-immunotherapy.
