Encapsulated cell therapy for local anti-CTLA-4 delivery in cancer immunotherapy.

Delivering cancer immunotherapy directly into tumors via an implanted device reduces life-threatening side effects while maintaining tumor-fighting power.

To address the dose-limiting systemic toxicities of anti-CTLA-4 (ipilimumab), Release Therapeutics developed Myo-P3—a retrievable encapsulated-cell device implanted peritumorally that delivers low, sustained anti-CTLA-4 doses directly into the tumor microenvironment. In a murine colorectal cancer model, Myo-P3 reduced intratumoral FoxP3+ Tregs and hCTLA-4+ CD4+ T cells (consistent with CTLA-4 blockade) with markedly lower systemic antibody exposure, supporting a potentially safer local immunotherapy platform.
