Combining DARTS proteomics, in vitro/in vivo mechanistic validation, and a prospective clinical trial, this study identifies GSK3β as a direct binding target of dapagliflozin in diabetic kidney disease podocyte protection. The rs60393216 GSK3B polymorphism was prospectively associated with individual UACR response, representing the first pharmacogenomic predictor of dapagliflozin renal efficacy.
