Glofitamab-Based Therapy for Relapsed or Refractory CNS Lymphoma: A Multicenter Retrospective Study

A new antibody drug achieves remission in most patients with relapsed brain lymphomas, with mostly mild side effects that doctors can manage.

In 84 R/R CNS lymphoma patients, glofitamab monotherapy achieved ORR 88% (CRR 59%) and combination therapy ORR 100% (CRR 81%) in PCNSL; median PFS was 19.5 months (PCNSL) and 13.5 months (SCNSL); CRS in 40% of patients was exclusively grade 1-2. This record was retained from the prior triage attempt for PubMed pipeline handoff.

Why it surfaced

Largest multicenter dataset for glofitamab in relapsed/refractory CNS lymphoma; response rates dramatically exceeding historical treatment options in a disease with near-zero durable salvage therapies. Retrospective design limits interpretation, but the effect size and consistency across PCNSL/SCNSL subtypes is compelling and likely to accelerate prospective trial design.
