Real-World Experience With TRBC1 Immunohistochemistry Across Cutaneous T-Cell Lymphoma Subtypes: A Large Cohort Study
A staining technique rivals molecular genetic testing for diagnosing skin T-cell lymphomas while costing less and being easier to implement in routine practice.
In a real-world validation cohort of 665 CTCL biopsies from 566 patients, TRBC1 immunohistochemistry achieved 85.8% sensitivity, 79.8% specificity, and 83.3% accuracy for T-cell clonality versus paired TCR sequencing. Digital QuPath quantification matched manual reads at 87.6% agreement, providing an implementable lower-cost alternative to molecular TCR testing for CTCL diagnostic workup.
