Molecular and immune profiling of HER2-low, HER2 ultra-low, and HER2-null male breast cancer.

Male breast cancer patients show distinct molecular features that might make them suitable for new antibody therapies approved for female breast cancer, potentially expanding treatment options.

This study provides the first molecular and immune profiling of male breast cancer stratified by HER2-low, ultra-low, and null categories, an important distinction given T-DXd's approval in HER2-low female breast cancer but lack of data in male breast cancer. The findings characterize whether male breast cancer patients may benefit from HER2-targeted ADC therapy and identify unique immunologic features that could inform treatment planning in this understudied, underserved population.
