Efficacy assessment in trials of complex and rare diseases: a comparison between the meta-analytic global statistical test and co-primary analysis.
This simulation study compared the meta-analytic global statistical test (GST) with co-primary analysis for assessing efficacy in trials of complex and rare diseases with multiple heterogeneous endpoints, finding the GST consistently demonstrated greater statistical power regardless of inter-outcome correlation. These results support consideration of the GST as an efficient primary analysis strategy in rare disease trials where traditional single-primary or co-primary approaches may be underpowered.
