Integrated analysis of tumor tissue and cfDNA sequencing across multiple DLBCL cohorts and TCGA identified 22 high-confidence driver genes and validated cfDNA as a complementary tool that captures core oncogenic signals while revealing heterogeneity unavailable from tissue alone. A four-gene expression model (MYC, PLCL1, IRF8, LNPEP) provided modest prognostic improvement over IPI.
